Pierwszych nagrań dokonał z orkiestrą Billy'ego Wrighta w 1951r. Pierwszy singel Taxi Blues i kilka następnych nie odniosły sukcesu. W Houston, w stanie Teksas, nagrywał z Johnny Otis Orchestra. W lutym 1955r przesłał próbne nagrania do wytwórni Speciality, która szybko zorganizowała artyście sesję nagrań w Nowym Orleanie. Rozpoczęła się kilkuletnia parada przebojów Little Richarda, najważniejsze z nich to: Tutti Frutti, Long Tall Sally, Rip It Up, Ready Teddy, Lucille, Keep A Knockin', Good Golly Miss Molly. Popularność artysty wzmocnił udział w filmach Don't Knock the Rock i Mr Rock and Roll.
W latach 1958-62 nagrywał muzykę gospel. W 1962r wystąpił po raz pierwszy w Europie, koncertując z The Beatles i The Rolling Stones. Z tego czasu pochodzi wielki przebój Bama Lama Bama Loo. Przez następne lata zmieniał wytwórnie, często występował w Las Vegas. W 1976 na dziesięć lat rzucił muzykę dla pracy duszpasterskiej.
W latach 80-tych z powodzeniem zagrał w filmie Down and Out in Beverly Hills. W 1986 został powołany w poczet Rock and Roll Hali of Fame. Potem często pełnił rolę gospodarza uroczystości show-biznesu, a w 1993 wystąpił podczas inauguracji prezydentury Billa Clintona. W 2000 koncertował w Europie wspólnie z Chuckiem Berrym i Jerrym Lee Lewisem.
