Po ukończeniu Chicago Musical College w wieku 16 lat Frank Skinner rozpoczął karierę w wodewilu wraz ze swoim bratem Carlem i rozpoczął długą karierę jako muzyk i aranżer zespołów tanecznych, głównie na parowcach pływających po rzece Illinois. Praca ta sprowadziła go pewnego dnia do Nowego Jorku, gdzie w latach 1925–1935 zaaranżował dla Robbins Publishing około 2000 popularnych piosenek. Przed wyjazdem z Manhattanu do Hollywood napisał dwie książki na temat tej dyscypliny, w tym New Method for Orchestra Scoring Skinnera (1935).
Po krótkiej pracy w MGM przy montażu muzycznym Wielkiego Ziegfelda (1936) Skinner został zatrudniony przez Universal Studios. W ciągu trzydziestu lat pracy w Universalu skomponował muzykę do ponad 200 filmów. Kontynuując pracę nad musicalami, szybko opanował specyficzne wymagania repertuaru dramatycznego i zdobył pięć nominacji do Oscara (1938-1943). Jego szczególne podejście do muzyki do filmów grozy, takich jak Syn Frankensteina (1939) i Człowiek-wilk ( 1941), charakteryzuje się „pasją do stylu chromatycznego… efektów lustrzanych… typowych orkiestracji, trzeźwości ale pełen grozy.
Nowe uznanie zyskał w latach pięćdziesiątych dzięki swoim ekstrawaganckim, romantycznym i melodramatycznym partyturom, zwłaszcza do Le Secret Magnificent (1954),Written on the Wind (1956) czy Imitation of Life (1959) Douglasa Sirka, które pozostają doskonałościami gatunku.
