Ostatnio jest też liczącym się wykonawcą popularnej muzyki chrześcijańskiej.
Przyszedł na świat w wielodzietnej rozinie (11 rodzeństwa), w której wszyscy muzykowali amatorsko. Pierwszą gitarę dostał od rodziców gdy miał 4 lata, jako nastolatek opuścił dom rodzinny i rozpoczął koncerty ze swoim wujem Dickiem Billem.
W 1960 przeniósł się do Los Angeles, gdzie szybko zdobył uznanie doskonałego muzyka sesyjnego i koncertowego. Współpracował wówczas z Frankiem Sinatrą (Strangers in the Night), Elvisem Presleyem, Rickiem Nelsonem, Johnnym Cashem, Deanem Martinem, Bobbym Darinem, Merle Haggardem (The Legend of Bonnie and Clyde), The Monkees (I'm a Believer), The Mamas and the Papas, Gene'em Clarkiem i z producentem muzycznym Philem Spectorem.
W 1965 przez prawie rok współpracował jako gitarzysta basowy - z zespołem The Beach Boys. Dla macierzystego wydawnictwa Capitol nagrywał piosenki, jednak dopiero jego wersja utworu Universal Soldier Buffy Saint-Marie przyniosła uznanie.
W 1967 nastąpił - za sprawą piosenki Gentle on My Mind autorstwa Johna Hartforda - przełom, utwór znalazł się na czołowych miejscach list przebojów zarówno country, jak i pop. Campbell stał się niezrównanym interpretatorem piosenek Jimma Webba (m.in. Wichita Lineman, Galveston, By the Time I Get to Phoenix).
Pod koniec lat 60. i w latach 70. wylansował liczne przeboje, m.in. Try a Little Kindness, Honey Come Back, Everything a Man Could Ever Need, It's Only Make Believe. Piosenki nagrywał w duecie z wokalistką Bobbie Gentry (Let It Be Me, All I Have to Do is Dream). Rozpoczął także udaną karierę aktorską, występując w filmach Johna Wayne'a. W 1969 został gospodarzem popularnego seryjnego programu telewizyjnego CBS The Glen Campbell Good Time Hour. W tym czasie osiągnął szczyt popularności.
Po kilkuletniej przerwie powrócił na pierwsze miejsca list przebojów za sprawą piosenki Rhinestone Cowboy, znanej z filmu Nocny kowboj. Ostatnim wielkim przebojem Campbella była piosenka Southern Nights, która dotarła do pierwszego miejsca list bestsellerów country i pop.
W połowie lat 80. wyszedł z uzależnienia od narkotyków i alkoholu, ponownie przyjął chrzest i skupił swoją energię na wielkiej trasie koncertowej, którą odbywał razem z Johnem Harfordem, Jimem Staffordem i Nicolette Larson, obejmującej ok. 200 występów rocznie. W połowie lat 90. poświęcił się grze w golfa i występom we własnym Goodtime Theater w Branson w stanie Missouri. W 1994 opublikował autobiografię Rhinestone Cowboy.
