We wczesnych nagraniach grupy Ross śpiewała tylko w chórkach. Główną wokalistką uczynił ją "pan i władca" Motown, Berry Gordy. Rolę tę miała spełniać w zespole przez sześć lat, przy czym płyty wydawane od 1967 r. sygnowane były nazwą Diana Ross And The Supremes.
U schyłku związku z grupą zadebiutowała równolegle jako solistka. Berry Gordy sprawował ścisły nadzór nad jej karierą, zaś plotka głosiła, że związek między artystką i jej opiekunem ma nie tylko muzyczny charakter. Pod koniec 1969 r. zapowiedziano rozstanie piosenkarki z The Supremes.
Wspólny pożegnalny koncert odbył się w styczniu 1970 r.
Pierwszym solowym bestsellerem Ross był singel "Ain't No Mountain High Enough" nagrany pod koniec tego samego roku. W kwietniu 1971 r. Diana poślubiła biznesmena Roberta Silbersteina. Małżeństwo przetrwało pięć lat, zaś rozwodowi towarzyszyła kolejna fala spekulacji na temat związków z Gordym.
Pod wpływem sukcesów typowo rozrywkowych singli solistki, wytwórnia postanowiła rozszerzyć jej estradowe emploi. W 1971 r. zadebiutowała we własnym programie telewizyjnym "Diana !". Rok później wystąpiła po raz pierwszy na ekranie w filmie "Lady Sings The Blues", biografii jazzowej śpiewaczki Billie Holiday w jej schyłkowym, narkomańskim okresie.
Późniejsze role w filmach "Mahogany" (1975 r.) i "The Wiz" (1978 r.) zostały przyjęte przez krytykę o wiele chłodniej (choć reżyserem drugiego z filmów był sam Sidney Lumet ).
W 1973 r. ukazał się wspólny album Diany Ross i Marvina Gaye'a, nagrany podobno oddzielnie i zmiksowany techniką studyjną. Tematy "Touch Me In The Morning" oraz motyw przewodni z "Mahogany" trafiły w 1975 r. na szczyt amerykańskich list przebojów. Singel "Love Hangover" z 1976 r. wykazywał wpływy muzyki disco, czego potwierdzeniem był album Diana wyprodukowany przez Nile'a Rodgersa i Bernarda Edwardsa z grupy Chic.
Ross była w tym czasie gwiazdą mass mediów, a informacje o jej romansie z Genem Simmonsem z zespołu Kiss wypełniały pierwsze strony gazet. Krążyły też pogłoski o dwuznacznych związkach z Michaelem Jacksonem, promowanym przez piosenkarkę jeszcze w latach 60.
Po licznych spekulacjach Ross odeszła ostatecznie z Motown w 1981 r., podpisując kontrakty z RCA na rynku amerykańskim i z Capitolem w pozostałych krajach. Założyła własną firmę producencką, a interpretacje tematów "Why Do Fools Fall In Love" Frankiego Lymona i "Muscles" Michaela Jacksona ugruntowały pozycję wokalistki w muzyce popowo-dyskotekowej.
W sierpniu 1981 r. na szczyt amerykańskiej listy przebojów weszła (utrzymując się tam przez 9 tygodni) nagrana wespół z Lionelem Richie'em piosenka "Endless Love", pochodząca z filmu Franka Zefirellego pod tym samym tytułem. Zarejestrowano ją podczas jednej krótkiej sesji w Reno ze względu na bardzo napięty rozkład zajęć obu gwiazd.
We wrześniu 1984 r. sporym powodzeniem cieszyła się nagrana w duecie z Julio Iglesiasem kompozycja "All Of You". Jednak jedynym nagraniem z końca łat 80. dorównującym popularnością dawnym przebojom był dedykowany pamięci Marvina Gaye'a utwór "Missing You".
Pociechą był brytyjski sukces singla "Chain Reaction" z 1986 r., autorstwa The Bee Gees (również przez nich wyprodukowany), naśladującego dawne brzmienie The Supremes.
W tym samym roku Ross poślubiła norweskiego stoczniowego potentata, kładąc tamę pogłoskom o związku z Gordym i rychłym powrocie do Motown. Późniejsze recitale Diany Ross (w tym głośny występ w nowojorskim Central Parku podczas ulewnego deszczu) dominowały zdecydowanie nad sporadycznymi nagraniami, lądującymi z reguły na odległych miejscach murzyńskich list przebojów.
Nagrany w 1991 r. album Force Behind The Power był płytą równie nudną, co mało ambitną i słusznie pozostał w cieniu wydanej dwa lata wcześniej kompilacji największych przebojów piosenkarki. Wczesną jesienią 1995 r. kolejny koncert artystki w Central Parku uświetnił premierę najnowszego przeboju wytwórni Disneya "Pocahontas".
