Był nominowany do Oscara za najlepszą piosenkę jedenaście razy i zdobył trzy Oscary za skomponowanie " Lullaby of Broadway ", " You'll Never Know " i "On the Atchison, Topeka and the Santa Fe ". Pisał muzykę do pierwszego kinowego musicalu filmowego, 42nd Street , w choreografii Busby Berkeley'a , z którym   współpracował przy wielu filmach muzycznych.
W ciągu kariery trwającej cztery dekady, Warren napisał ponad 800 piosenek. Inne znane hity Warrena to " I Only Have Eyes for You ", "You Must Have Been a Beautiful Baby ", "Jeepers Creepers ", "The Gold Diggers' Song (We're in the Money)", "That's Amore", "The More I See You", "At Last" i "Chattanooga Choo Choo" (z których ostatnia była pierwszą złotą płytą w historii).
W 1918 roku powołano go do U.S. Navy.
21 z nich osiągnęło  nr 1 na Your Hit Parade.Jego piosenka " I Only Have Eyes for You " jest wymieniona na liście 25 najczęściej wykonywanych utworów  20. wieku, opracowanej przez amerykańskie Stowarzyszenie Kompozytorów, Autorów i Wydawców ( ASCAP ).

Pierwszym przebojem Warrena był  utwór "Rose of the Rio Grande" (1922), z tekstem  Edgara Leslie .Napisał kolejne przeboje w latach 20-tych, w tym "I Love My Baby (My Baby Loves Me)", i "Seminola" w 1925, "Where Do You Work-a John?" ,"In My Gondola" w 1926 i "Nagasaki" w roku 1928. W 1930 roku skomponował muzykę do piosenki "Cheerful Little Earful" dla rewii  na Broadway'u ,Billy Rose  , Sweet and Low i skomponował muzykę do słów Morta Dixona i Joe Younga, dla    rewii  Eda Wynna The Laugh Parade z 1931 roku.

Przeniósł się do MGM, począwszy od 1944 roku, pisząc muzykę do filmów muzycznych , takich jak The Harvey Girls i The Barkleys of Broadway, wielu z udziałem Freda Astaire . Później pracował dla wytwórni Paramount , począwszy od początku lat 50-tych, pisząc do filmu Binga Crosby'ego Just for You   i  filmu Martina i Lewisa The Caddy , drugi zawierający przebój " To Amore ". Wciąż pisał piosenki do kilku kolejnych komedii Jerry Lewisa .

Warren zdobył Oscara za najlepszy utwór trzy razy, współpracując z trzema różnymi autorami tekstów : " Lullaby of Broadway " z Alem Dubinem w 1935 roku, "You'll Never Know " z Mackiem Gordonem w 1943 roku, oraz " On The Atchison, Topeka a Santa Fe " z Johnnym Mercerem w 1946 roku .Był nominowany do jedenastu Oscarów.
Warren poślubił Josephine Wensler w 1917r. Mieli syna, Harry Jr. (1919-1938), i córkę, Joan (ur. 1925). Jego żona zmarła w 1993 roku.

Warren zmarł 22 września 1981r w Los Angeles .  Jest pochowany w Westwood Village Memorial Park Cemetery w Los Angeles.
Teatr na Brooklynie  jest nazwany jego imieniem.
Warren mieszkał w Los Angeles i raczej unikał rozgłosu.
