Profesjonalną karierę zaczął w Chicago od współpracy (1937-39) w orkiestrze Earla Hinesa. Big-band Hinesa opuścił w 1943, by rozpocząć karierę solową. W latach 1944-47 stworzył (wraz z Buddym Johnsonem) własny big-band, lansujący nowoczesny bebop. Jak na owe czasy, była to jedna z najnowocześniejszych orkiestr jazzowych, w której grali wybitni innowatorzy modern jazzu i pionierzy bebopu: trębacze Dizzy Gillespie, Fats Navarro, Miles Davis i Kenny Dorham; saksofoniści Gene Ammons, Dexter Gordon, Sonny Stitt, Lucky Thompson, Charlie Parker, Leo Parker; pianista John Malachi; basista Tommy Potter i perkusista Art Blakey. Nagrania aranżowali: Budd Johnson, Tadd Dammeron i Jerry Valentine, natomiast wokalistką była Sarah Vaughan.
Billy Eckstine śpiewał i grał na puzonie wentylowym. Orkiestra nie zyskała wielkiej popularności, więc wokalista powrócił do zespołów swingowych, eksponując przede wszystkim swoją sztukę wokalną. W latach 50-tych Billy Eckstine stał się idolem amerykańskiej estrady (wraz z lukratywnym, jak na owe czasy, 5-letnim kontraktem płytowym z prestiżową MGM Records). Z jednej więc strony Eckstine był popularnym wokalistą i ckliwym balladzistą, z drugiej artystą, który w swoich zespołach lansował innowacje nowoczesnego jazzu.
