Ukończył zaledwie 4 lata, gdy cienkim głosikiem zaśpiewał Yes We Have No Bananas, i niewiele więcej, kiedy podjął naukę gry na fortepianie.
Jako dwunastolatek grał na organach kościelnych, lecz bardzo szybko zafascynował go jazz. Taki jazz, jaki w owym czasie grali Armstrong, Hines, Noone. Nathaniel Cole zorganizował swój pierwszy zespół, z którym w 1936 r. wystąpił w rewii Shuffle Along. O dalszej karierze zadecydował przypadek - niespodziewane wykonanie przeboju Noone'a Sweet Lorraine w jednym z klubów Hollywoodu. Publiczność natychmiast "kupiła" Cole'a i jego sposób interpretacji: nie minął rok, a Nat Cole Trio legitymowało się już całkiem interesującym kontraktem.
W 1940 r. Sweet Lorraine znalazła się na debiutanckiej płycie, której nalepkę okrasił dumny tytuł "King" przy nazwisku solisty. Tak też zostało, kiedy rok później Cole zaczął występować i nagrywać zawodowo. Ponieważ był przekonany, że mały zespół to niewielkie sukcesy, a sławę zapewni dopiero big-band - od 1948 r. nagrywał najchętniej z dużymi orkiestrami: Franka De Vola, Lesa Baxtera, Nelsona Riddle'a.
Chyba miał rację, skoro ponowny debiut przyniósł mu pierwszą Złotą Płytę za balladę Nature Boy. Na szczęście nie zaniechał Cole tak lubianego przez publiczność klubową śpiewania "od fortepianu" i wraz ze swym triem (z gitarzystą i kontrabasistą) nagrywał całkiem udane przeboje, jak choćby Little Girl w 1948 r. czy później Embraceable You, A Fool Was I, Gina, Route '66 i Non dimenticari oraz Dear Lonely Hearts i All Over the World.
Gdy latem 1962 r. obchodził jubileusz 25-lecia pracy artystycznej, fetowano go jak bohatera, odczytując publicznie gratulacje od prezydenta Kennedy'ego, ciesząc się z pamiątkowego złotego mikrofonu, ofiarowanego mu przez jego macierzystą firmę płytową Capitol. Warto pamiętać o tej powszechnej radości z dorobku czarnego śpiewaka pochodzącego z Mekki rasistów - Alabamy. To przecież w tym stanie sześć lat wcześniej atakowano Cole'a za jego pojawienie się na scenie, zarezerwowanej dla białych Hali Miejskiej w Birmingham, l wcale nie pomogła wtedy rekomendacja fachowego tygodnika "Billboard", że jedynie Nat "King" Cole i śpiewak country Webb Pierce gwarantują klientów w sklepie płytowym i publiczność w sali koncertowej.
Wielkich przebojów wylansował Cole sporo. Poza wymienionymi: Mona Lisa (z filmu Captain Carey, 1950), Too Young w 1951 r., Answer Me, My Love w 1954, A Blossom Fell i Darling (Je vous a/me beaucoup).
