Gladys Knight-urodziła się 28 maja 1944 roku w Atlancie/Georgia.Jest córką murzyńskich śpiewaków gospel związanych z chórem WINGS OVER JORDAN.Swoją pierwszą grupę założyła w rodzinnym mieście w 1952r.Jej skład stanowili głównie członkowie rodziny;brat "Merald" Bubba [ur.4.09.42r],jej siostra Brenda,kuzyni Elenor Guest i William Guest [ur. 2.06.41].Używali wówczas nazwy PIPS i śpiewali głównie gospels.Pierwsze nagrania zrealizowali dla wytwórni Brunswick w 1958 roku minn. SP "Whistle my love" już z nowymi członkami zespołu kuzynem Gladys-Edwardem Pattenem [ur. 2.08.1939r]i Langstonem George,którzy zastąpili wchodzących w związek małżeński Brendę i Elenora.
Na ich pierwszy hit "Every beat of my heart"[cover Johnny Otisa]wyprodukowany przez małą firmę Huntom trzeba było czekać trzy lata.W tym czasie zmieniają nazwę na Gladys Knight & The Pips i firmę płytową na Fury Rec [minn. reedycja "Every beat of my heart".
W 1964 roku mimo przenosin do Maxx Rec i wpółpracy ze znanym producentem Van MCCoye' m zaznacza się ten okres stagnacją w ich karierze.W 1964 r odchodzi Langston George i grupa występuje odtąd jako kwartet aż do lat 80-tych.
W 1966 roku następuje przełom w ich karierze dzięki współpracy z producentem i kompozytorem Normanem Whitfieldem i wytwórnią Soul [oddziałem Tamla Motown] dla której nagrywa całą serię przebojowych singli.
Na przełomie lat 60-tych i 70-tych grupa zmienia także za sprawą głównie rynku swój styl wnosząc więcej harmonii middle-of-the -road do swego oryginalnego rhythm'n' bluesowego brzmienia.
W 1972 opuszczają koncern Tamla dla Buddah niezadowoleni z pracy promocyjnej firmy z Detroit. Okazuje się ten krok posunięciem zbawiennym zwłaszcza dla ich popularności w Europie.Lata 70-te to przede wszystkim ich większa obecność w mediach;soundtrack dla filmu "Claudine" [1974],debiut aktorski Gladys w filmie "Pipedream", a nawet własny seryjny program TV [1975].
Po krótkim okresie współpracy w latach 80-tych z CBS grupa nagrywa dla MCA.pod koniec lat 80-tych grupa rozpada się a Gladys Knight sporadycznie nagrywa nowe solowe albumy a także udanie współpracuje z innymi artystami minn. Chaka Khan.
W 2001 roku wychodzi zamąż za długoletniego partnera życiowego Williama McDowella .
