Pierwszym nagranym dla niej singlem był pełen rozmachu "Reet Petite", który w USA poniósł porażkę, z trudem wchodząc na najniższe miejsca listy przebojów. Jednak w Wielkiej Brytanii trafił na 6. miejsce zestawień zapisując nazwisko Wilsona na trwałe w pamięci wielbicieli muzyki pop. Autorami tej piosenki byli Berry Gordy i Tyran Carlo (Roquel "Billy" Davis), którzy napisali dla Wilsona jeszcze kilka utworów, m.in. przebojowe "Lonely Teardrops" (z 1958 r.), "That's Why (I Love You So)" (z 1959 r.) i "I'll Be Satisfied" (z 1959 r.).
W 1960 r. Jackiemu udało się dwukrotnie wejść na szczyt rhythm'n'bluesowej listy przebojów z utworami "Doggin' Around" i "A Woman, A Lover, A Friend". Potem artysta wyraźnie nie mógł zdecydować się na muzyczny kierunek, który powinien obrać, miotając się pomiędzy muzyką środka a pseudo-operą. Udało mu się co prawda zabłysnąć w piosenkach takich, jak: "Baby Workout" (z 1963 r.), "Squeeze Her Please Her" (z 1964 r.) i "No Pity (In The Naked City)" (z 1965 r.), ale zbyt często jego aksamitny głos marnował się w przypadkowych, szybko wychodzących z mody utworach. Jednak w występach na żywo udało mu się utrzymać zarówno wysoki poziom, jak i dramatyzm, zdolny wprawić publiczność w ekstazę, czego często nie potrafiły dokonać jego pozbawione wyrazu nagrania.
Kariera Wilsona ożywiła się ponownie w 1966 r., kiedy to porzucił Nowy Jork przenosząc się do Chicago, gdzie nawiązał współpracę z producentem Carlem Davisem. Zarejestrowane tam "Whispers (Gettin' Louder)" (z 1966 r.), "(Your Love Keeps Lifting Me) Higher And Higher" (z 1967 r.) i wysublimowany "I Get The Sweetest Feeling" (1968 r.) należały do jego najbardziej udanych piosenek. Dobra passa nie trwała jednak długo. "This Love Is Real (I Can Feel Those Vibrations)" (z 1970 r.) okazał się ostatnim przebojem Wilsona z Top 10 rhythm'n'bluesowej listy przebojów, a on sam, zamiast ustanawiać nowe trendy, odzwierciedlał jedynie najpopularniejsze z nich w swoich piosenkach. 29 września 1975 r., w czasie trasy z rewią Dicka Clarka, na scenie Latin Casino w New Jersey doznał ciężkiego ataku serca. Padając na scenę uderzył się w głowę i w wyniku urazu mózgu wpadł w czteromiesięczną śpiączkę. W szpitalu przebywał z przerwami aż do śmierci 21.01.1984 r.
Jego kariera do dziś pozostaje zagadką. Pomimo początkowej współpracy i przyjaźni z Berrym Gordym, nigdy nie dołączył do jego imperium - wytwórni Motown. Pozostawione przez niego muzyczne dziedzictwo jest bardzo nierówne: momentami zachwyca, by zaraz rozczarować. W swoim utworze, "Jackie Wilson Said", uwiecznił go Van Morrison, a w wykonaniu formacji Dexy's Midnight Runners piosenka ta w 1982 r. dotarła do Top 5 brytyjskiej listy przebojów. Ironicznym zrządzeniem losu w 1987 r., dzięki nowatorskiemu wideoklipowi (który w opinii wielu obrażał pamięć wokalisty), wykorzystującemu animowane figurki z modeliny, utwór "Reel Petite" trafił na l. miejsce zestawień.
