Kwintet miał w repertuarze oryginalne interpretacje utworów
sakralnych, a jego brzmienie zdominowane było przez kontrastujący ze
sobą duet: mocny głos Mavis Staples i delikatniejszy tenor jej ojca.
Dodatkowym wyróżnikiem formacji była gra na gitarze Popsa zdradzająca
inklinacje bluesowe. Popularność przyniosły grupie single: "Uncloudy Day", "Will The Circle Be Unbroken" czy "I'm Coming Home". Utwór "This May Be The Last Time" zainspirował z kolei The Rollings Stones do napisania przeboju "The Last Time".
W pierwszej połowie lat 60-tych Staple Singers próbowali poszerzyć swój repertuar. Dwa single, których producentem był Larry Williams, "Why (Am I Treated So Bad)" i "For What It's Worth" - kompozycja Stephena Stillsa - wytyczały nowy kierunek, w którym zespół miał podążać po podpisaniu w 1967 r. kontraktu z wytwórnią Stax. Zaczął wtedy nagrywać kompozycje znanych autorów współpracujących z tą firmą, m. in. Homera Banksa i Bettye Crutcher, o tematyce raczej moralizatorskiej niż religijnej. Po odejściu Pervisa kwartet nagrał swoją wersję utworu Bobby'ego Blooma "Heavy Makes You Happy" (z 1970 r.), która była pierwszym rhythm'n'bluesowym hitem zespołu. Kolejne single "Respect Yourself (1971 r.), "I'll Take You There" (popowy hit numer l w USA w 1972 r.), "If You Are Ready (Come Go With Me)" (z 1973 r.) i "City In The Sky" (z 1974 r.) wzmocniły pozycję grupy na listach przebojów.
Staple Singers wzięli również udział w dwóch filmach, "Wattstax" i "Soul To Soul". Później kwartet przeniósł się do wytwórni Curtom i natychmiast odniósł sukces wykonując dwie filmowe kompozycje Curtisa Mayfielda "Let's Do It Again" (kolejny numer l w USA) i "New Orleans". Były one zarazem ostatnimi dużymi przebojami zespołu. W latach 1984-85 Staple Singers opublikowali kilka mniej udanych singli rhythm'n'bluesowych.
